Evaluation of a novel approach for measuring RNA quality
This poster relates to work carried out under project DD4 - Quality metrics and performance indicators for array based technology platforms of the NMS Chemical and Biological Metrology programme.
The purity and integrity of RNA samples are key determinants of quality of data generated in downstream assays such as microarrays.
Existing metrics frequently applied to RNA integrity assessment include 28S/18S rRNA ratio, RNA Integrity Number (RIN) determination using the Agilent Bioanalyser and 5’/3’ transcript signal intensity ratio determined by qRT-PCR or microarray analysis.
This poster describes the performance of a novel system for measuring RNA integrity, developed for use on the TapeStation® electrophoresis platform (Lab901). ScreenTape® R6K is a rapid separation matrix that allows resolution of RNA fragments. The resulting ScreenTape® Degradometer Value (SDV) gives a measure of the extent of degradation of the sample. The performance of the Lab901 TapeStation® platform was compared to that of the Agilent 2100 Bioanlyser and also SDV to RIN as a metric for the determination of RNA integrity when applied to microarray analysis.
This poster was presented at the 5th annual Advances in Microarray Technology (AMT) conference held in Stockholm, Sweden in May 2009.
