Analysis of Mono-phosphate Nucleotides Using High Performance Liquid Chromatography-Inductively Coupled Plasma Mass SpectrometryThis poster relates to work carried out under project S2 – Speciation of sulfur and phosphorus using chromatography and interference-reducing ICP-MS for pharmaceutical and industrial applications of the NMS Chemical and Biological Metrology programme.

Accurate quantification of DNA, nucleotides and nucleic acids is extremely important in biomedical research but is hindered by the lack of a reliable analytical technique and nucleic acid certified reference material (CRM).

Current analytical techniques, such as measuring the absorbance at 260nm and the use of fluorescence probes, are often compromised by impurities in the DNA. As phosphate forms the backbone of both RNA and DNA, and due to its fixed stoichiometry, the quantification of these macro-molecules can be achieved from the 31P signal by ICP-MS.

This poster was presented at the European Winter Conference on Plasma Spectrochemistry held in Graz, Austria in February 2009.
